I am not sure where the noise is coming from. I have a 96 disco with OME 2" HD lift.

While driving in neutral with the car off the noise comes from the back end. It sounds as if it might be coming from the Rotoflex. But I am not sure because it happens at all speeds not just while driving at higher speeds.

Does anyone know what this noise might be and more importantly how much will it cost me?

Bob:
First thing, climb beneath the truck and look for shiny metal indicating contact and wear. If your roto has won badly enough, the ears of the flanges could be striking each other.

Does it make a different noise when backing up?

It is probably time for you to be replacing the roto-flex and universals anyway on a seven year old truck, if you haven't already done it.

As for cost,(little finger in corner of mouth) One Million Dollars.

Peace,
Paul
